AntioxidantsplayanimportantroleinpreventingtheformationofandscavengingoffreerADIcalsandotherpotentiallytoxicoxidizingspecies.Therearethreecategoriesofantioxidantspecies:enzymesystems(GSHreductase,catalase,peroxidase,etc.),smallmolecules(ascorbate,uricacid,GSH,vitaminE,etc.)andproteins(albumin,transferrin,etc.).Differentantioxidantsvaryintheirreducingpower.Troloxisusedtostandardizeantioxidants,withallotherantioxidantsbeingmeasuredinTroloxequivalents.MeasurementofthecombinednonenzymaticantioxidantcapacityofbiologicalfluidsandothersamplesprovidesanindicationoftheoverallcapABIlitytocounteractreactiveoxygenspecies(ROS),resistoxidativedamageandcombatoxidativestress-relateddiseases.Insomecases,theantioxidantcontributionofproteinsisdesiredwhereasinothercasesonlythecontributionofthesmallmoleculeantioxidantsisneeded.HansaBioMeddevelopedtheTACAssayKit,whichcanmeasureeitherthecombinationofbothsmallmoleculeantioxidantsandproteinsorsmallmoleculesaloneinthepresenceofourproprietaryProteinMask.Cu²ᶧionisconvertedtoCu+bybothsmallmoleculeandprotein.TheProteinMaskpreventsCu²ᶧreductionbyprotein,enablingtheanalysisofonlythesmallmoleculeantioxidants.ThereducedCu+ionischelatedwithacolorimetricprobegivingabroadabsorbancepeakaround570nm,proportionaltothetotalantioxidantcapacity.
