Xanthineoxidase(XO,EC1.17.3.2)ispresentinappreciableamountsintheliverandjejunuminhealthyindividuals.However,invariousliverdisorders,XOisreleasedintocirculation.Therefore,determinationofserumXOlevelservesasasensitiveindicatorofacuteliverdamagesuchasjaundice.HansABIoMedhasdevelopedaneasyandsensitiveassaytodetermineXOinvarietyofsamples.Intheassay,XOoxidizesxanthinetohydrogenperoxide(H₂O₂)whichreactsstoichiometricallywithOxiRed™Probetogeneratecolor(atλ=570nm)andfluorescence(atEx/Em=535/587nm).SincethecolororfluorescenceintensityisproportionaltoXOcontent,theXOactivitycanbeaccuratelymeasured.Thekitdetects1-100mUxanthineoxidasein100µlreactionvolume.
