Description:

Myeloperoxidase(MPO)isaperoxidaseenzyme(EC1.11.1.7)mostabundantlypresentinneutrophilgranulocytes.Itisagreenhemoproteinfoundinneutrophilsandmonocytesthatcatalyzesthereactionofhydrogenperoxideandhalideionstoformcytotoxicacidsandotherintermediatesthatplayaroleintheoxygen-dependentkillingoftumorcellsandmicroorganisms.Itshemepigmentcausesthegreencolorinsecretionsrichinneutrophils,suchaspusandsomeformsofmucus.FurThermore,itcanoxidizetyrosinetoatyrosylrADIcalusinghydrogenperoxideasanoxidizingagent.IntheMPOAssayKit,MPOcatalyzestheproductionofNaClOfromH₂O₂andNaCl.Subsequently,NaClOwillreactstoichiometricallywithAminophenylfluorescein(APF)togeneratefluorescein,whichhasastrongfluorescenceandcanbedetectedatEx/Em=485/525nm.Thekitprovidesarapid,simple,sensitive,andreliabletestsuitableasahighthroughputassayofMPOactivity.ThiskitcanbeusedtodetectMPOactivityaslowas0.5μUperwell.
