Description:
Glutathione(GSH)isthemajorintracellularlow-molecular-weightthiolthatplaysacriticalroleinthecellulardefenseagainstoxidativestressinmammaliancells.HansABIoMed’sApoGSH™GlutathioneColorimetricAssayKitprovidesaconvenient,colorimetricmethodforanalyzingeithertotalglutathioneorthereducedformglutathionealoneusingamicrotiterplatereader.TheassayisbasedontheglutathionerecyclingsystembyDTNBandglutathionereductase(Fig.1).DTNBandglutathione(GSH)reacttogenerate2-nitro-5-thiobenzoicacidwhichhasyellowcolor.Therefore,GSHconcentrationcanbedeterminedbymeasuringabsorbanceat412nm.ThegeneratedGSSGcanbereducedbacktoGSHbyglutathionereductase,andGSHreactswithDTNBagaintoproducemore2-nitro-5-thiobenzoicacid.Therefore,therecyclingsystemdramaticallyimprovesthesensitivityoftotalglutathionedetection.Thekitincludesthe5-Sulfosalicylicacid(SSA)fortheremovalofproteinsfromsamplesandfortheprotectionofGSHoxidationandγ-glutamyltranspeptidasereaction.Thekitcanquantifyglutathionefrom1-100ng/wellina200lreaction.Fordetectinglowerglutathioneconcentrations,suchasinbloodsamples,increasingreactiontimewillgeneratestrongersignal.Thekitcanalsospecificallydetectthereducedformofglutathione(GSH)byomittingtheglutathionereductasefromthereactionmixture.Thesensitivityfordetectingthereducedformofglutathione(withoutrecyclingsystem)is100timeslowerthandetectingthetotalglutahione.
